I love history questions "Yahoobunny427!"
Ok, well at first glance it would seem a simple question, however there is more to it than you might think.
Most historians agree that the first retail establishment was the Roman forum. After all, people went there to barter for goods.
However it was in nearby Babylon that retail fronts were organized for the purpose of taxing and consumer safety.
Many of these rules were laid out in the ancient Code of Hammurabi. If a retail establishment occupied more than 1000 Babylonian cubits (about 530 mm or 20.9 inches) it was called a Estoreabrium. If it took up less than 1000 Babylonian cubits it was called an Ashoporium. The larger establishment paid a slightly higher tax (or tribute) and they were allowed to sell food items. The smaller paid less tax but could only sell non-edible goods.
The terms Estoreabrium and Ashoporium were used for close to 1000 years, but when Europe started to add retail establishments at the end of the Dark Ages they shortened the names to "Store" and "Shop." These store and shop owners became the first middle class. They were above the subsistence farmer but below the nobles of the day. Truly in "the middle."
Fast forward to 2024 and there is no real difference. I suppose most folks would still think of shops as being smaller and more specialized, and stores as being larger and sometimes part of a chain.
